N2 - The aim of the study was to investigate the content of chondroitin sulfate (CS) in neurons and the extracellular matrix of different layers of the neocortex of SCID mice treated with temozolomide (TMZ) and in modeling the growth of glial tumor. Material and methods. The work was carried out on male SCID mice (n = 24) aged 10 weeks weighing 20–25 g. TMZ at a dosage of 30 mg/kg in a volume of 240 μl was administered intragastrically for 3 cycles of 5 days in a row. To obtain experimental tumors, mice were orthotopically injected with suspensions of U87 glioblastoma cells into the subcortical structures of the brain. The content of CS in coronary sections of the brain was studied by the immunohistochemical method, the results of the reaction were assessed using a score system. Results. TMZ administration to intact mice resulted in a decrease in the CS content in the cells of the molecular, external granular, pyramidal, and internal granular layers and a decrease in the CS content in the extracellular matrix of the molecular and external granular layers (p < 0.05). In the absence of TMZ, the grafted glial tumor did not affect the CS content in the cells and extracellular matrix of the neocortex. In contrast, TMZ administration to mice with tumor was accompanied by a decrease in the CS content in the cells of both the external granular and pyramidal layers of the neocortex, whereas in the extracellular matrix it decreased only in the external granular layer. Conclusions. TMZ reduces the CS content in the cerebral cortex of experimental animals regardless of the presence or absence of a grafted glial tumor, but the degree of expression of the recorded changes is higher in the intact brain and varies depending on the layer of the neocortex. Since TMZ affects the studied parameter in the layers that play a key role in the implementation of cognitive functions, one of the reasons for the impairment of these functions in neuro-oncological patients may be long-term use of TMZ and the heterogeneous degree of the drug’s effect on a particular cellular layer of the neocortex.
