N2 - Background: The creation of effective psychotropic drugs is a key problem of psychopharmacology. Anxiolytic [1], anticataleptic [2] and antidepressant [3] effects of 8-(trifluoromethyl)-1,2,3,4,5-benzopentathiepin-6-amine hydrochloride (TC-2153), an inhibitor of striatal enriched tyrosine phosphatase (STEP), on mice were shown. Zebrafish (D. rerio) is a common object in preclinical studies of new psychotropic drugs. The aim of the study was to investigate the effects of acute and chronic TC-2153 treatment on the nervous system and behavior of adult zebrafishes. Method(s): The experiments were carried out on 3-4 months old males and females (50:50) of long fin zebrafishes. For acute treatment, fishes were isolated for 3 h in tanks (0.5 l) containing vehicle (0.05% DMSO and 0.05% Tween 20), 0.125 mg/l or 0.25 mg/l of TC-2153. For chronic treatment, fishes were kept in groups of 12 in aerated tanks (4 l) containing vehicle or 0.125 mg/l of TC-2153. The vehicle and drug solutions were changed every day for 14 days then the fishes were isolated for 3 h in 0.5 l tanks containing corresponding solutions. Behavior of the fishes was tested in the "novel tank" (NT) using EthoStudio software, then the fishes were euthanized in ice cold water and the monoamines concentrations in the brains were measured by HPLC. In the experiment with manganese enhanced MRI (MEMRI) the fishes were placed for 1 h in tanks (0.5 l) containing 10 mM MnCl2 together with vehicle, 0.125, 0.25 or 0.5 mg/l of TC-2153, then the fishes were euthanized and placed in 15 ml falcon tubes containing 10 ml of 2% agarose for MRI. The density of accumulated Mn2+ was evaluated as the rate of MRT signal in the telencephalon to MRT signal in reference image.
